在免疫反应过程中,抗原加工细胞将静止的B细胞活化为浆细胞,以分泌针对该抗原的免疫球蛋白。从多个参与免疫反应的浆细胞克隆中,可以通过普通的细胞培养技术将每个分离。一旦分离,该细胞及其后代就会产生相同亚型的相同免疫球蛋白。这些免疫球蛋白于是被称为单克隆抗体,并且对所需抗原的一个表位具有相同的亲和力和特异性。
